Injection of sgRNA + CAS9 complex before pronuclei have faded implies editing occurs during S and G2 phase (before cytokinesis) and is thought to minimize mosaiscism.
Injection of sgRNA + CAS9 after pronuclear fading implies editing occurs in G1 phase of the 2 cell stge (after cytokynesis) and is thought to promote mosaicism.
Targeting OCT4 reduces survival of embryos after EGA; significantly less embryos reach blastocyst stage; blastocyst quality is compromised.
Suggestion: OCT4 has a role in early human preimplantation development.
Indel analysis showed a high frequency of 2‐3 bp deletions, although larger indels were also observed. Off‐target effect not observed.
