 The HPV Vaccine‐ Intervention
• HPV‐infection is cleared naturally in approximately 90% of cases
• The L1 capsid protein contains multiple overlapping immunodiominant epitopes, However, in natural HPV infection the anti‐ L1 humoral immune response is weak and type‐specific. And though neutralizing is delayed and is not detected consistently following infection.
• Yet, the correlate for protection seems to be anti‐L1 humoral responses :
• The recombinant Vaccine s(L1 capsid protein) resembles the conformation of
authentic virions; are neither infectious, nor oncogenic;
• induce high levels of type specific neutralizing antibodies (HOW)?
• The antigen dose in VLPs is much higher than in natural infection and the capsids are directly exposed to systemic immune responses. VLPs are highly immunogenic [two log over the natural infection], inducing high concentrations of neutralizing Ab to L1 AND generate a heterogeneous or polyclonal immune response:
• Microabrasions during the infection of basal membrane allow exudation of these neturalizing antibodies thus blocking the life cycle of the virus
