 Learning Objectives
Chromosomal instability in human pluripotent stem cells:
• Type and frequency of chromosomal abnormalities in hiPSCs and hESCs • Mechanisms leading to chromosomal instability hPSCs
• Culture heterogeneity
• Exogenous effects on chromosomal stability
• Implications for clinical applications
Most abnormal hPSCs are chromosomally mosaic in culture
• International Stem Cell Initiative-> most abnormal cultures were a mosaic of genetically divergent cells
• Mosaic aneuploidy also observed in cells of the blastocyst inner cell mass from which hESCs are derived
• Cell to cell variability may also lead to that observed in hESCs
• Both hESC and somatic cells exhibit highly mosaic cultures
