  Human testicular organoids: an approach to reconstitute in vitro a testis-like organ

 •Isolation of human SSCs
•Immortalization of Sertoli and Leydig cells •Preparation of human testis extracellular matrix •Formation of testis organoids
•In vitro culture for 23 days in hanging drops •Evaluation of the organoid organization by
immunofluorescence and PCR analysis of different cell type specific markers (SCP3, PRM1, FSHr etc)

Advantage: determine the gonadotoxicity of drugs
Limitation: very low efficiency of germ cell differentiation
 CONCLUSIONS
• In vitro production of functional sperm from gonocytes/SSCs is feasible in the mouse
• Among different in vitro models presently available, the most efficient is the organ culture in a pumpless microfluidic system. Using neonatal mouse testis, this model maintains the two essential functions of the male gonad, spermatogenesis and steroidogenesis, for 6 months
• The somatic gonadal environment is required for a successful in vitro male germ cell differentiation
