Diagnostic tools: 2. Mouse Oocyte Calcium Analysis (MOCA)
Allows the study of the pattern of the Ca2+ oscillations induced by human sperm upon ICSI in a mouse model.
Method:
• Patient’s sperm is microinjected into mouse
oocytes pre‐loaded with a Ca2+ indicator.
• The analysis of parameters such as the peak amplitude (A) and frequency (F) are used to
define the score AxF. The score AxF describes a threshold for normality at the value of >9 a.u.

• MII mouse oocytes were loaded with the calcium indicator Fura‐2AM (7.5μM) (for 20 min in the incubator and 10 min at room temperature)
• ICSI of frozen/thawed test or donor sperm: piezo‐ICSI (15‐17°C)
• Calcium measurement for 2 hours (37°C, 6%CO2)
• +‐15 oscillating oocytes per group
